To increase the effectiveness of the drug and lose weight, it is necessary to control the diet and avoid excessive fat intake.

Daily fat intake should be evenly distributed over three main meals, because if orlistat is taken with a very high-fat meal, the risk of gastrointestinal complications may increase.

Orlistat reduces the absorption of some fat-soluble nutrients, especially beta-carotene and vitamin E. Therefore, the diet should be balanced in terms of food and rich in fruits and vegetables.

Orlistat may reduce the absorption of vitamins needed by the body, so taking a multivitamin is recommended during the course of taking the medication.

In patients with diabetes, weight loss with orlistat may lead to improvement in the disease and the attending physician may change the dose of oral diabetes medications or insulin.

Orlistat prevents fat absorption, so this medicine should be taken with food and 1 hour after it.

Dosage and method of use of the medicine:

The amount of medicine is determined by the attending physician for each patient. However, the usual dosage of the medicine is as follows:

For the treatment of obesity

Orlistat is the most widely used anti-obesity drug with the fewest side effects. This drug has been introduced as a weight loss, weight maintenance, and weight gain prevention drug by potently and reversiblely inhibiting pancreatic lipase and preventing the absorption of dietary fat. Unlike other weight loss drugs such as phentermine, which reduces appetite by stimulating the central nervous system; Orlistat acts locally in the gastrointestinal tract and reduces the absorption of dietary fat by up to 30%.

Orlistat is used as a treatment for obesity, and it is expected that a person will lose 5 to 10% of their initial weight or 5 to 9 kilograms per year. At the same time, the response of individuals to Orlistat is not the same; for example, in people with type 2 diabetes, the weight loss process is slower and requires long-term use of the drug. On the other hand, in these people, due to the risk of mortality from macrovascular problems, in addition to weight loss, improving the cardiovascular profile such as blood lipids and blood pressure and improving lifestyle are also considered therapeutic goals. Studies have shown that taking orlistat for 6 to 12 months causes a significant reduction in blood cholesterol.

Absorption: The absorption of the drug after oral administration is very low and reaches maximum plasma concentration within 6 to 8 hours. The onset of action of orlistat is 24 hours after administration and the bioavailability of the drug is reported to be 5%.

Metabolism: Orlistat is metabolized in the intestinal wall and produces inactive metabolites M1 and M3.

Excretion: 95 to 97% of orlistat is excreted in the feces and less than 3% in the urine. The elimination half-life of the drug is about 1 to 2 hours.

General Interaction Profile:
– Decreased absorption of fat-soluble substances
• Use caution when co-administering with oral medications with a narrow therapeutic window. Orlistat may decrease the absorption of some oral medications.
